چکیده

The ability to detect surfaces within confocal microscope images is important for the interpretation of such data. Most detectors are gradient-based, and so are sensitive to noise, and fail to detect some feature types that are important in confocal microscopy. The local energy feature detector developed by Morrone and Owens marks locations where there is congruence of phase in the Fourier components of an image. Points of maximal phase congruency occur at all common feature types: step and roof edges, line features and Mach bands. A 3D implementation of the local energy feature detector, suitable for confocal microscope data, is presented. The detector computes local energy by convolving an image with oriented, quadrature pairs of 3D lters. To increase the speed of the convolution, the lters are designed in frequency space and multiplied by the image's Fourier transform. Some preliminary results are presented.

Feature Extraction of Chromosomes from 3D Confocal Microscope Images

An investigation of local energy surface detection integrated with neural network techniques for image segmentation is presented, as applied in the feature extraction of chromosomes from image datasets obtained using an experimental confocal microscope. Use of the confocal microscope enables biologists to observe dividing cells (living or preserved) within a three-dimensional (3-D) volume, that...

3D Confocal Scanning Laser Microscopy to Quantify Contact Angles in Natural Oil-Water Mixtures

3D Confocal Scanning Laser Microscopy to Quantify Contact Angles in Natural Oil-Water Mixtures — The contact angle of a liquid drop on a solid surface is one of the most simple, useful and sensitive parameters in the hydraulic sciences to quantify and to qualify the wettability and the surface energy of different materials. In this paper, a confocal scanning laser microscope (CSLM) has been use...

3D Fourier analysis methods for digital processing and 3D visualization of confocal transmission images

We have developed digital 3D Fourier transform methods for comparing the 3D spatial frequency content and hence the axial and transverse resolution of confocal versus conventional microscope images. In particular, we have utilized these techniques to evaluate the performance of our recently-developed confocal transmission microscope for bright field and Nomarski DIC imaging. We have also found ...

A Reaction-diffusion Algorithm for Segmentation of Confocal Laser Scanning Microscope Images

The images obtained using a confocal laser scanning microscope (CLSM) alone cannot detect threedimensional (3D) structures, for example of the endothelial cells that compose blood vessels, and require segmentation for 3D interpretation. Here, we investigated the segmentation of hepatic sinusiodal veins, which form periodic 3D networks. We propose a new approach for image segmentation based on t...

Real-time True-color Volume Visualization of Multi-channel 3D CLSM Images Based on CUDA

Confocal laser scanning microscope (CLSM) can obtain multichannel 3D images from biological tissues for analysis of tissue structures, where each channel results from a particular fluorescent label. The visualization of the multi-channel 3D confocal microscope images is required for qualitative, interactive, and even quantitative analysis of the structures of the studied tissues. Despite the av...
